1305 Involucrin/ck1e/vitamin D receptor regulatory axis underlies evolution of human epidermis out of Africa

چکیده
The human epidermis has evolved out of Africa. Increased involucrin (IVL) expression in the skin is associated with a near selective sweep Northern European populations compared to African. significance for this positive selection not clear. Our functional studies using Ivl-/- mice recently determined requirement positively regulate Vitamin D receptor and its activity epidermis. mouse treated MC903 (vitamin agonist) exhibited dampened Vdr response notable decreased expressions multiple scRNA-seq-identified keratinocyte populations. To further ascertain mechanism by which dosage impacts activity, we employed comprehensive multi-omics approach (ATAC-seq, RNA-seq, LC/MS proteomics) determine chromatin accessibility, transcriptomic, proteomic changes WT newborn (q<0.05). Csnk1e (casein kinase 1e) emerged as top candidate at intersect these differential datasets less accessible 3’UTR coinciding isoform reduced peptide protein (PK) domain observed skin. findings reveal an emerging genomic regulatory axis Ivl/Ck1e/Vdr keratinocytes. We next African ancestral epidermis, marked relatively lower IVL expression, decrease CK1e isoforms PK domains VDR higher expression. Together, our mouse-to-human translational identify conserved Ivl/CK1e/VDR that underlies evolution highlights population-specific, molecular diversity

Primate evolution — in and out of Africa
A synthetic analysis of molecular, fossil and biogeographical data gives a remarkably consistent scenario for the evolution of the catarrhine primates - the hominoids and Old World monkeys. This analysis supports the African location of the common ancestor of the Old World monkeys, and suggests that the Asian colobine monkeys and macaques dispersed out of Africa into Eurasia within the past ten...
متن کاملHuman origins: Out of Africa
Our species, Homo sapiens, is highly autapomorphic (uniquely derived) among hominids in the structure of its skull and postcranial skeleton. It is also sharply distinguished from other organisms by its unique symbolic mode of cognition. The fossil and archaeological records combine to show fairly clearly that our physical and cognitive attributes both first appeared in Africa, but at different ...

The serial founder model of modern human origins predicts that the phylogeny of ancestries exhibits bifurcating, tree-like behavior. Here, we tested this prediction using three methods designed to investigate gene flow in autosome-wide genotype data from 3,528 unrelated individuals from 163 global samples. Specifically, we investigated whether Cushitic ancestry has an East African or Middle Eas...
متن کاملHuman evolution out of Africa: the role of refugia and climate change.
Although an African origin of the modern human species is generally accepted, the evolutionary processes involved in the speciation, geographical spread, and eventual extinction of archaic humans outside of Africa are much debated. An additional complexity has been the recent evidence of limited interbreeding between modern humans and the Neandertals and Denisovans. Modern human migrations and ...
